Asterand Expanding Service To Include Expression Profiling From FFPE Samples


Asterand plc, a certified service provider utilizing NuGEN's products, is a leading global supplier of high quality, well characterized human tissue and human tissue-based research services to drug discovery scientists. The company provides human tissues and services to accelerate the identification and validation of therapeutic targets and enhance the selection of drug candidates for its clients.

As interest has grown in linking the molecular basis of disease to the clinical outcomes of patients, Asterand's clients have been increasingly drawn to utilizing specimens from donors where the treatment and therapeutic outcomes are known. Asterand has an extensive biorepository of materials with clinical outcomes data. However, the majority of these samples are in an FFPE format which can impact the ability to recover intact RNA suitable for quantitative RT-PCR studies. Asterand had utilized several commercial kits for recovering intact RNA from these tissues with little success. In 2008, the company approached NuGEN® to incorporate its Whole Transcriptome WT-Ovation RNA amplification kit into its PhaseZERO® Gene Expression service platform.

Asterand now utilizes the kit on its CustomMapping™ product line that offers clients a platform for exploring diseased and normal gene expression using quantitative RT-PCR and specimens from its biorepository. The kit has performed admirably within the lab personnel's hands. As Dr. Samir Alhasan, Director of Cell and Molecular Services at Asterand's Detroit facility notes "NuGEN's Ovation RNA amplification kit literally rescued a difficult FFPE project we performed for a client in 2008. The laser capture micro-dissection (LCM) of the client's FFPE tumor yielded very few cells for study. The Ovation kit allowed us to recover and amplify sufficient RNA to complete quantitative RT-PCR successfully." The success of this project led the company to incorporate the kits into its platform and seek NuGEN certified provider status.

Publications HSF1-Mediated BAG3 Expression Attenuates Apoptosis In 4-Hydroxynonenal Treated Colon Cancer Cells Via Stabilization Of Antiapoptotic Bcl-2 Proteins

Summary
Heat shock factor 1 (HSF1) promotes cytoprotective heat shock gene expression, and silencing HSF1 sensitizes cells to 4-Hydroxynonenal (HNE) induced apoptosis. The authors of this study used Affymetrix GeneChip 1.0 ST arrays to investigate the regulation of the Bcl-2 gene family by HSF1. This gene family is of interest due to the established roles of its members in an array of diseases including arterial restenosis, rheumatoid arthritis, hepatic carcinoma, multiple myeloma, prostate cancer, and colon cancer.

In the gene expression portion of this study, HSF1 was silenced by siRNA transfection, and control and silenced RNA samples from normal colon and colon cancer cell lines were amplified in triplicate using the WT-Ovation FFPE RNA Amplification System V2. Sense target was generated with the WT-Ovation Exon module and fragmented and labeled with the FL-Ovation cDNA Biotin module. Results showed that BAG3, a protein co-chaperone, is regulated by HSF1. BAG3 expression significantly attenuates HNE-mediated apoptosis in colon cancer cells.

Silencing HSF1 results in diminished expression of BAG3. The decreased expression of BAG3 in turn leads to diminished levels of Bcl-XL, Mcl-1, and Bcl-2 protein expression, however, the mRNA levels of these genes were not affected by knockdown of HSF1 or BAG3, suggesting that the effect is mediated at the level of protein turnover. BAG3 has also been shown to inhibit the degradation of Hsp70 client proteins, and the authors propose that the cytoprotective mechanism of BAG3 involves the stabilization of the anti-apoptotic Bcl-2 family members in a multi-protein complex with BAG3 and Hsp70.
