The Encore SP+ Complete DR Multiplex Systems provide a completely automated end-to-end solution for strand-specific RNA-Seq library construction using as little as 100 ng of total RNA on the Mondrian™ SP+ Workstation. The core technology used in this enriches for non-ribosomal mRNA in NGS libraries during first strand cDNA synthesis and can be applied to transcriptomes extracted from a broad range of higher eukaryotes. No dedicated steps are required for rRNA reduction. Proprietary primers are used to create double-stranded cDNA which retains RNA strand information. In a single process without user intervention, the resulting cDNA is converted to an NGS library suitable for analysis on any Illumina sequencing platform. The Encore SP+ Complete DR Multiplex Systems provide optional barcodes for multiplexed applications.
The streamlined workflow consists of the following steps:
1. Addition of total RNA and reagents to the Mondrian SP+ Cartridge
2. Hands-free automation of the following assay steps on the Mondrian SP+ Workstation:
- Sample concentration
- 1st and 2nd strand cDNA synthesis
- Sample purification
- Enzymatic fragmentation
- Sample purification
- End repair
- Sample purification
- Adaptor ligation
- Sample purification
- Library enrichment
- Enriched library purification
3. Collection of the final enriched, purified library droplet from the cartridge
4. The following steps are conducted separately from the Mondrian SP+ Workstation:
- Quantification
- Cluster formation and sequencing (with optional sample multiplexing)
The Encore SP+ Complete protocol can be completed in 10 hours with less than 30 minutes of hands-on time - a 90% reduction in hands-on time compared to the manual Encore SP+ Complete protocol.
The Encore SP+ Complete DR Multiplex System 1-8 (Part No. 8151) and Encore SP+ Complete DR Multiplex System 9-16 (Part No. 8152) both provide eight unique barcoded adaptors for multiplex sequencing. Used in combination, these two kits enable up to 16-plex sequencing using a dedicated read (DR) barcode design.
