Mondrian SP Applications

Library Preparation — Method 1
This method automates the process of constructing DNA sequence libraries in the Mondrian SP Universal Cartridge (PN 8010) using NEBNext® DNA Sample Prep library reagents on the Mondrian™ SP digital microfluidics system. The protocol requires 100 ng of fragmented DNA as input and produces indexed or non-indexed libraries ready for quantitation and normalization prior to cluster generation and analysis on any Illumina sequencing-by-synthesis platform.

Application Note #1: Mondrian SP Library Preparation Method 1

Coming Soon. The following protocols are currently in development and will be made available to Mondrian SP users in the near future.

Ovation® SP Ultralow DNA Library System
The Ovation® SP Ultralow Library System will provide an automated, fast and scalable solution for producing libraries starting with as little as 1.0 ng of DNA. The resulting libraries will be suitable for a wide range of sequencing applications on Illumina sequencing-by-synthesis platforms, including RNA-Seq, Digital Gene Expression (DGE), genomic DNA/exome sequencing, amplicon sequencing, ChIP-Seq and more. The method will be able to produce indexed and non-indexed libraries.

Encore® SP Complete RNA-Seq Library System
The Encore® SP Complete RNA-Seq Library Systems will provide a completely automated, end-to-end solution for strand-specific RNA-Seq library construction using as little as 100 ng of total RNA. The core technology used in this product will enrich for non-ribosomal mRNA in NGS libraries during cDNA synthesis, and can be applied to transcriptomes extracted from a broad range of higher eukaryotes. The first strand cDNA synthesis will be carried out using proprietary primers to create double-stranded cDNA which retains RNA strand information. No dedicated steps will be required to reduce rRNA levels. In a single process without user intervention, the resulting cDNA will be converted to NGS libraries suitable for analysis on any Illumina sequencing-by-synthesis platform. The Encore Complete RNA-Seq Multiplex Systems provide optional indexing.

Encore® Rapid DNA Library System
The Encore® Rapid DNA Library System will enable automated NGS library construction that eliminates the need for any amplifications steps. Using input of 100-500ng of genomic DNA or double stranded cDNA, the hands-free protocol will be completed within 2 hours to produce libraries ready for quantitation prior to cluster generation and analysis on any Illumina sequencing-by-synthesis platform. The method will be able to produce indexed and non-indexed libraries.
