Ovation® 3'-DGE System
The Ovation 3'-DGE System provides accurate expression analysis (tag profiling) suitable for quantification of any poly(A)+ transcripts, without the need for prior sequence knowledge. It employs a simple, fast, and automatable workflow, powered by the Ribo-SPIA Single Primer Isothermal Amplification technology and is compatible with all leading NGS platforms.
The input for the Ovation 3'-DGE System is flexible. You can choose one of the two options that best fits with your experimental design:
- 10 ng – 100 ng of purified total RNA
- Direct cell lysate of 50 – 10,000 cells using the Prelude™ Direct Lysis Module
The protocol for the Ovation 3'-DGE System takes only six hours to complete and generates micrograms of double-stranded cDNA, focusing on the most 3' 150 – 500 bases proximal to the poly(A) tail of the transcripts, ready for NGS library construction.
In particular, for users choosing the Illumina NGS platforms, the integrated solution with the Encore NGS Library System I or the Encore NGS Multiplex System I, DGE libraries can be produced in nine hours, greatly simplifying the entire sample preparation procedure. The combination of the Ovation 3'-DGE System and the Encore NGS Library System I provides a complete, streamlined and easily automatable sample preparation workflow, enabling NGS-based expression profiling approaching the protocol simplicity and sample throughput of microarrays.
